No, and this is the most common mistake with them. They arrive at the correct moisture content and are ready to use as supplied. Soaking waterlogs the plug and pushes out the air that roots need, which is a reliable way to rot a cutting. If a plug feels dry after sitting for a long time, mist it lightly rather than soaking.
Not at the start. Each plug carries a starter charge of nutrient, which is enough to carry a cutting or seedling through rooting. Adding feed to a plant with no root system just raises the salt level around it. Begin light feeding once roots are visible at the sides of the plug, not before.
The Plug & Grow trays are the plugs themselves in 12, 24 and 77 cube formats. The Sea of Green set and the Six Plug sowing set are packaged combinations aimed at a specific way of growing. When roots are visible emerging from the sides and base of the cube — that means the plug is colonised and the plant is ready for more room. Leaving it much longer starts to circle roots inside the cube, which slows establishment after potting on. The whole plug goes into the next container; there is no need to remove it.
