Yes, if you want the meter accurate across the range you actually grow in. Two-point calibration sets 7.0 (neutral) then 4.0, so the instrument reads true across the whole band growers use. A single-point check only proves the meter near that one number. That is why Growth Technology and Flairform sell the two buffers as a pair.
Once it is out of the bottle, treat it as single-use: calibrate, then tip it out. Do not dunk a dirty probe in the bottle, and do not pour used solution back — either will move the reference value. Unopened bottles keep. Aquamaster's 20mL sachets dodge the question entirely because each sachet is a fresh pour.
No. KCl is what you store in. Buffer holds a known pH, but it does not have the electrolyte concentration a glass electrode needs to stay conditioned. Put a few drops of KCl storage solution in the cap. Never leave the probe dry, and never sit it in distilled or RO water.
